Indications
Vitamin D3 deficiency may arise due to limited sunlight exposure or an inadequate diet. Vital for calcium and phosphate absorption, Vitamin D3 supports healthy bones and teeth, preventing conditions like rickets, osteomalacia, and osteoporosis. It’s also crucial for preventing pre-eclampsia during pregnancy and breastfeeding, serving as an essential nutrient for infants and boosting immunity.
Pharmacology
Calcitriol, the active form of Vitamin D3, binds to Vitamin D receptors (VDRs) distributed throughout body tissues. With a half-life of approximately 50 days, Vitamin D3 is fat-soluble and absorbed in the small intestine. It undergoes metabolism to 25-hydroxy Vitamin D3 (Calcidiol) in the liver and then to 1,25-dihydroxy Vitamin D3 (Calcitriol) in the kidney, facilitating calcium absorption and storage in tissues.
Dosage & Administration
- For Capsules (Adults): Treatment of Vitamin D3 deficiency: 40000 IU once weekly for 7 weeks. Maintenance therapy: 1400-2000 IU/day. Prevention: 20000 IU every 4 weeks. Addition to osteoporosis therapy: 20000 IU once a month.
- For Capsules (Children 12-18 years): Treatment: 20000 IU once every 2 weeks for 6 weeks. Prevention: 20000 IU every 6 weeks.
- For Film-Coated Tablets: 1000 IU (1-2 tablets) daily, with food or within 1 hour after a meal.
- For Oroflash or Chewable Tablets: 1000 IU to 2000 IU daily, with food or within 1 hour after a meal. Place the tablet in the mouth and swallow after chewing.
- For Syrup: Dosage varies based on age and deficiency risk.
Interaction
Certain drugs, including Magnesium-containing Antacids, Digoxin, and Corticosteroids, can cause Vitamin D3 deficiencies by interfering with absorption and metabolism.
Contraindications
Vitamin D3 is contraindicated in diseases associated with hypercalcemia and hypersensitivity to Vitamin D3 or its excipients.
Side Effects
Rare symptoms include anorexia, nausea, vomiting, diarrhea, constipation, weight loss, polyuria, sweating, headache, thirst, vertigo, and elevated plasma and urine calcium and phosphate concentrations.
Pregnancy & Lactation
Safe use during pregnancy up to 4000 IU/day has been observed. Breastfeeding mothers should consult a healthcare practitioner before additional supplementation.
