Indications
Vardamate is prescribed for the treatment of erectile dysfunction (ED).
Pharmacology
Vardenafil, the active ingredient in Vardamate, selectively inhibits phosphodiesterase type 5 (PDE5), thereby enhancing penile erection. It works by increasing the levels of cyclic guanosine monophosphate (cGMP), which promotes smooth muscle relaxation in the corpus cavernosum, leading to improved blood flow and erection during sexual stimulation.
Dosage & Administration
The recommended initial dose of Vardamate is 10 mg, taken orally approximately 60 minutes before sexual activity. Based on effectiveness and tolerance, the dose can be adjusted to a maximum of 20 mg or reduced to 5 mg. It is advised not to exceed one dose per day. Vardenafil can be taken with or without food.
For elderly patients (>65 years), a starting dose of 5 mg is recommended. No dose adjustment is necessary for patients with renal impairment. In patients with moderate hepatic impairment, a starting dose of 5 mg is advised, with a maximum dose of 10 mg. Vardamate is not recommended for patients with severe hepatic impairment.
Interactions
Vardamate may enhance the blood pressure-lowering effects of antihypertensive medications. Caution is advised when co-administering with other vasodilators or alpha blockers. It does not potentiate the hypotensive effect of alcohol. Certain medications such as ketoconazole, indinavir, ritonavir, and erythromycin may affect the excretion of Vardenafil from the body.
Contraindications
Vardenafil should not be used with nitrates or nitric oxide donors due to the risk of potentiated hypotensive effects. It is contraindicated in individuals with hypersensitivity to Vardenafil or any of its components.
Side Effects
Common side effects of Vardamate include headache, flushing, nasal congestion, dyspepsia, stomach upset, dizziness, and back pain.
Pregnancy & Lactation
Vardenafil is not indicated for use in women.
Precautions & Warnings
Physicians should evaluate the cardiovascular status of patients before prescribing Vardamate, considering the cardiac risks associated with sexual activity. It is not recommended for men with underlying cardiovascular conditions for whom sexual activity is contraindicated.
Overdose Effects
Human data on Vardamate overdose are limited. In cases of overdose, characterized by symptoms such as blurred vision and back pain, medical attention should be sought immediately.
