Indications
Ucol is prescribed for managing symptoms of overactive bladder, including urge urinary incontinence, urgency, and frequent urination.
Pharmacology
Tolterodine, the active ingredient in Ucol, acts as a competitive antagonist at muscarinic receptors, primarily targeting the bladder with minimal impact on salivary glands. It is metabolized into an active form that enhances its therapeutic effects, typically noticeable within four weeks of treatment.
Dosage & Administration
For extended-release capsules, the recommended initial dose is Tolterodine Tartrate 4 mg daily, adjusted based on individual response. Patients with hepatic or renal impairment or those taking potent CYP3A4 inhibitors should start with Tolterodine Tartrate 2 mg daily.
Film-coated tablets are generally prescribed as Tolterodine Tartrate 2 mg twice daily, adjusted to Tolterodine Tartrate 1 mg twice daily for patients with severe liver or kidney impairment. Dosage adjustments may be made based on tolerance.
Pediatric & Geriatric Use
Tolterodine’s safety and efficacy have not been established in children. However, no significant safety differences were noted between younger and older adult patients.
Interactions
Co-administration with potent CYP3A4 inhibitors like Ketoconazole may necessitate a lower Ucol dosage to mitigate increased plasma concentrations.
Contraindications
Ucol is contraindicated in patients with urinary retention, gastric retention, uncontrolled narrow-angle glaucoma, or hypersensitivity to Tolterodine Tartrate.
Side Effects
