Pharmacological Action
Rosuvastatin acts as a selective and competitive inhibitor of HMG-CoA reductase. This enzyme is pivotal in converting 3-hydroxy-3-methylglutaryl coenzyme A to mevalonate, a precursor of cholesterol. Dosage Information

For patients with HoFH, the starting dose is 20 mg/day. In pediatric patients with HeFH, the recommended dosage is 5-10 mg/day for those aged 8 to less than 10 years, and 5-20 mg/day for patients aged 10 to 17 years. Pediatric patients with HoFH should take 20 mg/day if aged 7 to 17 years.
Administration

Interactions

- Cyclosporine: Combination increases Rosuva exposure, limiting the Rosuva dose to 5 mg once daily.

- Lopinavir/Ritonavir or atazanavir/ritonavir: Combination increases Rosuva exposure, requiring the Rosuva dose to be limited to 10 mg once daily.
- Coumarin anticoagulants: Combination prolongs international normalized ratio (INR). Stable INR should be achieved before starting Rosuva, with frequent monitoring thereafter.
Contraindications
Rosuvastatin is contraindicated in individuals with:
- Known hypersensitivity to product components
- Liver disease, including unexplained persistent elevations in hepatic transaminase levels
- Pregnant or breastfeeding women
Side Effects
Rosuva is generally well-tolerated. Common adverse events associated with Rosuva include headache, myalgia, constipation, asthenia, abdominal pain, and nausea.
