Pyridox

Indications

Cyclid is indicated for the treatment of nausea and vomiting associated with pregnancy in women who have not responded to conservative management strategies.

Pharmacology

Doxylamine Succinate is an antihistamine that effectively blocks H1 receptors, which helps alleviate nausea. It crosses the blood-brain barrier, targeting H1 receptors in the brain to reduce the activity of histamine. Additionally, it inhibits the vestibular system and muscarinic receptors, contributing to its antiemetic effects. Pyridoxine Hydrochloride, a vitamin B6 analog, complements this action by further preventing nausea and vomiting.

Dosage & Administration

For initial treatment, take one tablet orally at bedtime on Day 1. If symptoms are well managed the following day, continue with one tablet daily at bedtime. If symptoms persist on Day 2, increase the dose to one tablet in the morning and one at bedtime. The maximum recommended dosage is two tablets per day, taken on an empty stomach with a glass of water. Swallow tablets whole; do not crush, chew, or split them. This medication should be taken daily rather than on an as-needed basis.

Interaction

Doxylamine Succinate and Pyridoxine Hydrochloride should not be used in conjunction with monoamine oxidase inhibitors (MAOIs), as this can amplify central nervous system side effects. The combination with alcohol and other CNS depressants is also not advisable.

Contraindications

This medication is contraindicated in women who have:

• A known hypersensitivity to Doxylamine Succinate, Pyridoxine Hydrochloride, or any inactive ingredients.
• Concurrent use of monoamine oxidase inhibitors (MAOIs), which can intensify adverse effects.

Side Effects

Common side effects may include drowsiness and other complications from interaction with CNS depressants.

Pregnancy & Lactation

Classified as pregnancy category A, this medication is intended for treating nausea and vomiting in pregnant women who do not respond to conservative measures. Both Doxylamine Succinate and Pyridoxine Hydrochloride are excreted in breast milk, so caution is advised during breastfeeding.

Precautions & Warnings

Doxylamine Succinate may cause drowsiness; avoid driving or operating heavy machinery while taking this medication. Caution is warranted for women using CNS depressants, including alcohol. This medication should also be used carefully in individuals with asthma, glaucoma, peptic ulcers, or urinary obstructions.

Overdose Effects

Overdose symptoms may not appear immediately due to the extended-release formulation. Signs of overdose include restlessness, dry mouth, dilated pupils, dizziness, confusion, and increased heart rate. Severe cases can lead to anticholinergic effects, seizures, or even death. Treatment may involve gastric lavage, activated charcoal, or symptomatic management.
