Perkirol is recommended for alleviating the symptoms associated with idiopathic Parkinson’s disease and also serves as an effective treatment for moderate-to-severe primary Restless Legs Syndrome (RLS).
Pharmacology
Ropinirole, a non-ergoline dopamine agonist, exhibits its highest affinity at the D3 receptors, primarily located in the limbic areas of the brain. While its precise mechanism of action for Parkinson’s disease remains elusive, it is believed to selectively stimulate dopamine D2 receptors within the caudate-putamen system, influencing bodily movement. Ropinirole’s impact on Restless Legs Syndrome is similarly attributed to its ability to activate dopamine receptors.
Dosage & Administration
For Parkinson’s disease, the recommended starting dose is 0.25 mg three times daily, with gradual titration up to a maximum daily dose of 24 mg. In the case of Restless Legs Syndrome, the initial adult dose is 0.25 mg once daily, increasing to a maximum of 4 mg daily over the course of a week.
Interaction
It’s important to note that dopamine antagonists like neuroleptics or metoclopramide may diminish Perkirol’s effectiveness due to its dopamine agonist properties.
Contraindications
Perkirol should not be administered to patients with known hypersensitivity to Ropinirole.
