Indications:
- Osetron is a serotonin subtype 3 (5-HT3) receptor antagonist used for:
- Preventing nausea and vomiting caused by chemotherapy (both initial and repeat courses).
- Preventing and treating post-operative nausea and vomiting (PONV).
- Preventing nausea and vomiting induced by radiotherapy.
Always use under the supervision of a registered healthcare provider.
Description:
Osetron Oral Soluble Film is an easy-to-use, dissolving film that is placed on the tongue, where it dissolves in 20 seconds, offering quick relief from nausea and vomiting. It doesn’t require water for swallowing. The active ingredient, Osetron base, works by blocking serotonin receptors, preventing nausea and vomiting induced by chemotherapy, radiotherapy, or surgery.
Pharmacology:
Osetron, a potent 5HT3 receptor antagonist, blocks the serotonin receptors in both the peripheral and central nervous system, reducing nausea and vomiting caused by chemotherapy, radiotherapy, or post-surgery. While the exact mechanism in post-operative nausea and vomiting is not fully understood, it likely shares similar pathways with chemotherapy-induced nausea.
Dosage:
Chemotherapy-Induced Nausea and Vomiting:
- Adults & Pediatric (6 months – 18 years): Three doses of 0.15 mg/kg (maximum 16 mg per dose), up to three doses for intravenous use.
Radiotherapy-Induced Nausea and Vomiting:
- Adults: 8 mg orally 1–2 hours before treatment and 8 mg every 8 hours for up to 5 days after radiotherapy.
Post-operative Nausea and Vomiting:
- Adults: 16 mg (two 8 mg doses) 1 hour before anesthesia.
- Pediatrics (40 kg or more): 4 mg injection.
Administration:
Osetron Oral Soluble Film:
- Step 1: Tear open the pouch.
- Step 2: Place the film on your tongue and allow it to dissolve within 20 seconds.
- Step 3: Do not chew; swallow once dissolved.
- Step 4: Wash hands after use.
Side Effects:
Common side effects of Osetron include headache, constipation, diarrhea, and in some cases, rash. Severe reactions may include anaphylaxis, tachycardia, and shortness of breath. Any unusual symptoms should be reported to a doctor.
Pregnancy & Lactation:
- Osetron is not known to have carcinogenic effects, but use during pregnancy should be carefully considered due to lack of well-controlled studies.
- Osetron is excreted in rat breast milk, so caution is advised when administered to nursing mothers.
Precautions & Warnings:
- Avoid use in patients with known hypersensitivity to 5-HT3 receptor antagonists.
- Osetron does not stimulate gastric or intestinal peristalsis and should not replace nasogastric suction.
- Use cautiously in patients with a history of hypersensitivity reactions or severe hepatic impairment.
Therapeutic Class:
- Anti-emetic drugs.
Storage Conditions:
- Store at a temperature not exceeding 30ºC. Protect from light and moisture.
