Description

Indications

Loratin tablet swiftly relieves symptoms associated with seasonal allergic rhinitis, perennial allergic rhinitis, and skin allergies like chronic urticaria. It effectively eases symptoms such as sneezing, nasal discharge, itching, ocular itching, and burning linked with allergic rhinitis. Administered orally, it promptly alleviates nasal and ocular signs and symptoms. Loratin tablet is also recommended for idiopathic urticaria. In children aged 2 and above, Loratin tablet provides symptomatic relief for seasonal allergic rhinitis and allergic skin conditions like urticaria and nettlerash.

Pharmacology

This tablet contains Loratadine, a non-sedative histamine H1 receptor antagonist with antiallergic properties. Loratadine, a long-acting tricyclic antihistamine, selectively antagonizes peripheral H1 receptors without inducing sedation or anticholinergic effects. Its rapid onset and prolonged action enable once-daily dosing.

Dosage & Administration

For adults and children over 12 years, one Loratadine tablet is recommended daily, preferably in the morning or as needed for symptom relief. For children aged 2-12 years:

  • Body weight over 30 kg: one Loratadine tablet daily
  • Body weight below 30 kg: half Loratadine tablet daily Loratadine tablet is not advisable for children below 2 years due to insufficient safety and efficacy data.

Interaction

No hazardous interactions with other drugs have been reported. Unlike many other H1 receptor antagonists, Loratin does not potentiate the effects of alcohol on psychomotor performance. However, concomitant use with drugs metabolized by hepatic cytochromes P450 3A4 and 2D6 may increase plasma concentrations, potentially leading to adverse effects.

Contraindications

Loratadine should not be used in patients with known hypersensitivity or idiosyncrasy to its components.

Side Effects

In controlled clinical studies, adverse events with 10 mg Loratin were comparable to placebo, with minimal sedation and anticholinergic effects reported. Studies on driving performance and cognitive functioning indicate no significant difference from placebo.

Pregnancy & Lactation

Due to limited data, Loratadine is not recommended during pregnancy. It is excreted in breast milk in negligible amounts, advising caution in nursing mothers.

Precautions & Warnings

Exercise caution in patients with liver impairment or renal insufficiency (eGFR <30 ml/min).

Overdose Effects

Overdose may lead to somnolence, tachycardia, and headache in adults, and extrapyramidal signs and palpitations in children. Prompt symptomatic and supportive measures should be initiated, including oral activated charcoal or gastric lavage if necessary.
