Description
Geston, an oral Gestagen, stands out for its potent pregnancy-maintaining action in castrated animals, devoid of hormonal side-effects, particularly the masculinization of female fetuses. Clinical investigations have underscored Geston’s ability to stimulate the endocrine function of the trophoblast. Moreover, in vitro studies demonstrate its capacity to enhance progesterone production in the placenta. Notably, Geston administration has shown promising results in restoring abnormally low levels of placental hormones and specific enzymes in patients with high-risk pregnancies. Most importantly, Geston therapy has proven effective in averting abortion threats and ensuring uncomplicated pregnancies. With its excellent tolerability, Geston is associated with minimal side-effects, if any.
Pharmacology
Allylestrenol, the active component of Geston, exhibits a mild progestational effect on the human endometrium. Notably, in vitro studies highlight its role in stimulating progesterone synthesis in the human placenta, along with an increase in specific placental enzymes. Histological analyses reveal enhanced placental activity following allylestrenol treatment, contributing to the maintenance of normal pregnancies. Clinical studies have further demonstrated allylestrenol’s ability to suppress uterine contractions without adverse effects on delivery progress. Importantly, allylestrenol exhibits no estrogenic or androgenic properties and does not impact adrenal function.
Dosage & Administration
For different conditions:
- Intra Uterine Growth Retardation: Take 1 tablet thrice daily for at least two months.
- Threatened abortion: Administer 1 tablet thrice daily until symptoms resolve.
- Habitual abortion: Begin with 1-2 tablets daily upon pregnancy confirmation, continuing for at least one month post the critical period.
- Threatened premature delivery: Individual dosage determination required, with high dosages (up to 40 mg daily) used in some cases.
In case of missed doses, take the medication as soon as remembered, without doubling the dose.

Contraindications
Avoid Geston in cases of:
- Breast cancer or related conditions
- Severe liver diseases
- Undiagnosed vaginal bleeding
- Toxaemia of pregnancy

Side Effects
Long-term Geston use may lead to gastrointestinal discomfort, including vomiting, nausea, and occasional epigastric discomfort.

Precautions & Warnings
