Description

Indications

Dinogest is prescribed for managing endometriosis. It belongs to the progestin class of medications, which mitigate the effects of estrogen on the endometrium (uterine lining) and breast tissue. By reducing estrogen’s influence, Dinogest alleviates pelvic pain associated with endometriosis.

Pharmacology

Dinogest, a progestin with strong effects on the endometrium, functions as a selective progesterone receptor agonist. It induces endometrial atrophy, reduces endogenous estradiol levels, and counteracts estrogen’s effects on both normal and ectopic endometrial tissues. It also alleviates androgenic symptoms like acne and hirsutism.

Dosage & Administration

  • Tablets: Start any day of the menstrual cycle. Take 2 mg daily without breaks, ideally at the same time each day. Continue without interruption and replace missed doses with an additional 2 mg.
  • Missed Doses: If a tablet is missed, take 2 mg as soon as possible and resume normal dosing the next day. Replace doses missed due to vomiting or diarrhea with an additional 2 mg.

Interactions

Dinogest is metabolized by CYP3A4. Drugs that affect CYP3A4 levels, such as certain antifungals, antibiotics, and protease inhibitors, may alter Dinogest’s effectiveness.

Contraindications

Avoid Dinogest in individuals with hypersensitivity to the medication or its components, active venous thromboembolic disorders, severe cardiovascular disease, diabetes with vascular issues, severe hepatic disease, liver tumors, hormone-dependent malignancies, or undiagnosed vaginal bleeding.

Side Effects

Common side effects include headache, breast discomfort, mood changes, and acne. Less frequent effects may involve gastrointestinal disturbances, weight increase, and mood alterations.

Pregnancy & Lactation

  • Pregnancy: Not recommended as its safety during pregnancy is not established.
  • Lactation: Dinogest is excreted in breast milk. Consider discontinuing breastfeeding or the medication based on the benefit-risk assessment.
