Vitamin D3 deficiency may arise in individuals with limited sunlight exposure or inadequate dietary intake. Vital for calcium and phosphate absorption, Vitamin D3 plays a crucial role in maintaining healthy bones and teeth, preventing conditions like rickets, osteomalacia, and osteoporosis. It’s also essential for preventing pre-eclampsia during pregnancy and breastfeeding and has a significant impact on boosting immunity.
Pharmacology
Calcitriol, the active form of Vitamin D3, binds to Vitamin D receptors (VDRs) distributed across various body tissues. Absorbed in the small intestine, Vitamin D3 undergoes metabolization to 25-hydroxy Vitamin D3 (Calcidiol) in the liver and further conversion to 1,25-dihydroxy Vitamin D3 (Calcitriol) in the kidneys, enhancing calcium absorption. Non-metabolized Vitamin D3 is stored in tissues like fat and muscle before elimination through feces and urine.
Dosage & Administration
Capsule:
- For adults: Treatment of deficiency – 40000 IU weekly for 7 weeks, followed by maintenance therapy of 1400-2000 IU/day. Prevention – 20000 IU every 4 weeks.
- For children (12-18 years): Treatment – 20000 IU once every 2 weeks for 6 weeks, Prevention – 20000 IU every 6 weeks.
- For film-coated/tablet/chewable tablet: 1000-2000 IU daily.
- For syrup: Dosage varies based on age and deficiency risk.
Interaction
Various drugs can lead to Vitamin D3 deficiency, including corticosteroids and antiepileptics, by interfering with absorption and metabolism.
Contraindications
Contraindicated in hypercalcemia, hypersensitivity to Vitamin D3, or evidence of Vitamin D3 toxicity.
