Pharmacology
Olopatadine, an antihistamine, works by blocking histamine, which helps reduce allergy symptoms. It effectively addresses sneezing, itching, and nasal congestion caused by allergies.
- Mechanism of Action:
Olopatadine stabilizes mast cells, preventing histamine release, and acts as a selective H1 receptor antagonist, reducing inflammatory mediators like prostaglandins and cytokines. - Pharmacokinetics:
- Absorption: Peak plasma concentrations occur 30 minutes to 1 hour after intranasal use.
- Distribution: Olopatadine binds moderately to proteins (about 55%).
- Metabolism: Minimal metabolism with six minor plasma metabolites.
- Elimination: Half-life is 8–12 hours, mainly excreted via urine.

Interactions
No significant drug interactions are anticipated, as olopatadine is primarily eliminated via renal excretion. Interactions related to P450 enzymes or plasma protein binding are unlikely.
Contraindications
Do not use if you are hypersensitive to any components of this product. Inform your doctor of any ongoing medications or health conditions before use.
Side Effects
Common side effects include headaches, blurred vision, dry eyes, or nasal discomfort. Rarely, allergic reactions or systemic effects like nausea and fatigue may occur.
Use During Pregnancy & Lactation
While animal studies show no teratogenic effects, adequate studies in pregnant women are lacking. Use only if benefits outweigh risks. Olopatadine’s excretion in human breast milk is unknown, so caution is advised for nursing mothers.
