Description

Clarizol is a versatile antifungal medication designed to combat dermal infections caused by a variety of pathogenic dermatophytes, yeasts, moulds, and bacteroids. In addition to its antifungal properties, Clarizol is effective against specific gram-positive bacteria, particularly Staphylococcus species, and Trichomonas. As with other imidazole compounds, Clarizol works by altering the permeability of fungal cell walls, disrupting their lipid structure. At lower concentrations, it primarily inhibits ergosterol synthesis, while at higher concentrations, it can induce additional damage to the fungal membrane independent of sterol synthesis.

Pharmacology

Clotrimazole, the active ingredient in Clarizol, operates mainly by compromising the cell membrane’s permeability barrier in fungi. It inhibits ergosterol biosynthesis, which is crucial for maintaining functional fungal cell membranes. When ergosterol production is fully or partially disrupted, the fungal cell loses its ability to form a complete and functional membrane. Since ergosterol plays a critical role in promoting fungal cell growth in a hormone-like manner, the onset of these changes results in a dose-dependent reduction in fungal proliferation.

Although the decrease in ergosterol due to the inhibition of lanosterol 14-demethylase (CYP51) is recognized as the primary mechanism behind clotrimazole’s antifungal effects, the drug also exhibits additional pharmacological properties. These include the inhibition of sarcoplasmic reticulum Ca²⁺ ATPase, depletion of intracellular calcium, and blockage of calcium-dependent potassium channels and voltage-dependent calcium channels, contributing to effects beyond its antifungal action.

Dosage & Administration

Clotrimazole Cream: Apply a thin layer to the infected area 2-3 times daily, gently massaging it in. Due to its high efficacy, a small amount typically suffices for an area roughly the size of the palm. Consistent and prolonged use of Clotrimazole Cream is vital for successful treatment. Treatment duration varies based on the severity and location of the infection.

Recommended Treatment Duration:

  • Dermatomycoses: 3-4 weeks
  • Candida vulvitis and Candida balanitis: 1-2 weeks
  • Erythrasma and Pityriasis Versicolor: Approximately 3 weeks

In cases of fungal infections on the feet, continue treatment for an additional 2 weeks following the complete resolution of symptoms to prevent relapse. Ensure feet are thoroughly dried after washing, particularly between the toes. Clotrimazole Cream is odorless, easily washable, and does not stain clothing.

Clotrimazole Topical Solution: Apply sparingly to the affected areas and rub in gently, 2-3 times daily.
