Pharmacology
Trypan Blue acts as a selective tissue stain (dye) specifically targeting the anterior lens capsule of the human crystalline lens. This dye does not penetrate the capsule, enabling clear visualization of the anterior capsule against the non-stained lens cortex and inner lens material.
Dosage & Administration
To administer Trypan Blue, begin by injecting an air bubble into the anterior chamber using a 26-gauge needle. This step prevents the dilution of the dye by aqueous humor. Next, use a tuberculin syringe to inject Trypan Blue into the anterior lens capsule. Staining is considered adequate once the dye contacts the capsule. If any area remains unstained, apply an additional drop until complete coverage is achieved.

Contraindications
Trypan Blue should not be used when a non-hydrated (dry state) hydrophilic acrylic intraocular lens (IOL) is intended for insertion, as the dye may be absorbed by the IOL, leading to discoloration.
Side Effects
- Discoloration of intraocular lenses with high water content.
- Unintentional staining of the posterior lens capsule and vitreous face.
- These effects are generally self-limiting and can last up to one week.
Pregnancy & Lactation
- Pregnancy Category C: There are no sufficient and well-controlled studies in pregnant women. Trypan Blue should only be administered to pregnant individuals if the potential benefits outweigh any potential risks to the fetus.
- Lactation: It is unclear whether Trypan Blue is excreted in human breast milk. Due to the possibility of many drugs being excreted in milk, caution is advised when administering Trypan Blue to nursing mothers.
Precautions & Warnings
After injection, it is crucial to thoroughly irrigate the anterior chamber to remove any excess Visi Blue immediately.
Use in Special Populations
- Pediatric Use: The safety and efficacy of Visi Blue have been confirmed in pediatric patients, supported by evidence from adequately controlled studies.
- Geriatric Use: No significant differences in safety or efficacy have been observed between elderly patients and younger individuals.
