Description

Indications

Perkirol is recommended for alleviating the symptoms associated with idiopathic Parkinson’s disease and also serves as an effective treatment for moderate-to-severe primary Restless Legs Syndrome (RLS).

Pharmacology

Ropinirole, a non-ergoline dopamine agonist, exhibits its highest affinity at the D3 receptors, primarily located in the limbic areas of the brain. While its precise mechanism of action for Parkinson’s disease remains elusive, it is believed to selectively stimulate dopamine D2 receptors within the caudate-putamen system, influencing bodily movement. Ropinirole’s impact on Restless Legs Syndrome is similarly attributed to its ability to activate dopamine receptors.

Dosage & Administration

For Parkinson’s disease, the recommended starting dose is 0.25 mg three times daily, with gradual titration up to a maximum daily dose of 24 mg. In the case of Restless Legs Syndrome, the initial adult dose is 0.25 mg once daily, increasing to a maximum of 4 mg daily over the course of a week.

Interaction

It’s important to note that dopamine antagonists like neuroleptics or metoclopramide may diminish Perkirol’s effectiveness due to its dopamine agonist properties.

Contraindications

Perkirol should not be administered to patients with known hypersensitivity to Ropinirole.

Side Effects

Common side effects of Perkirol encompass fainting, drowsiness, hallucinations, dizziness, nausea, vomiting, uncontrolled movements, leg swelling, fatigue, confusion, headache, upset stomach, abdominal discomfort, increased sweating, among others.

Pregnancy & Lactation

Perkirol falls under pregnancy category C, with limited studies on pregnant women. Its use during pregnancy should be considered only if the benefits outweigh potential risks. Additionally, Ropinirole’s inhibition of prolactin secretion may impact lactation.

Precautions & Warnings

Patients should be aware of the possibility of experiencing drowsiness leading to inadvertent sleep during daily activities. Moreover, the systemic regulation of blood pressure may be affected by dopamine agonists, potentially resulting in orthostatic hypotension.

Therapeutic Class

Perkirol belongs to the category of antiparkinson drugs.

Storage Conditions

Store Perkirol in a cool, dry place below 25°C, away from light and moisture, and ensure it is kept out of reach of children.
