Indications for Use
Patadin DS Eye Drops are designed to alleviate the discomfort associated with allergic conjunctivitis. This includes symptoms such as itchy, watery, red, and swollen eyes or eyelids. Patadin is effective for conditions such as vernal keratoconjunctivitis, vernal keratitis, blepharitis, blepharoconjunctivitis, and giant papillary conjunctivitis.
Olopan DS Nasal Spray is an antihistamine that provides relief from seasonal allergic rhinitis symptoms in both adults and children aged 6 years and older.
Olopan DS Tablets are used to manage allergic rhinitis (inflammation of nasal membranes due to allergens like pollen and mold spores) and allergic skin conditions. They help alleviate symptoms such as skin rashes, swelling, hives, itching, sneezing, and nasal congestion or runny nose.
Pharmacology
Olopatadine, the active ingredient, is an antihistamine that blocks histamine’s action, which helps reduce allergy symptoms. This medication effectively manages sneezing, itching, and nasal discharge caused by allergies.
Mechanism of Action: Olopatadine works as an anti-allergic agent through multiple pathways. It stabilizes mast cells and prevents the release of histamine and other inflammatory mediators. By antagonizing H1 receptors with high specificity, it blocks inflammatory pathways and inhibits the release of cytokines and other pro-inflammatory substances.
Pharmacokinetics:
- Absorption: Peak plasma levels of olopatadine are typically reached within 30 minutes to 1 hour after intranasal administration.
- Distribution: The drug binds moderately to serum proteins (~55%) and is not significantly affected by varying concentrations.
- Metabolism: Olopatadine undergoes minimal metabolism, with at least six minor metabolites present in the plasma.
- Elimination: The elimination half-life ranges from 8 to 12 hours, with the majority of the drug excreted through urine.
Dosage
Eye Drops:
- 0.1% Solution: One drop in each affected eye twice daily (every 6-8 hours).
- 0.2% Solution: One drop in the affected eye once daily.
- 0.7% Solution: One drop in each affected eye once daily.
Nasal Spray:
- Adults and Adolescents (12+ years): Two sprays per nostril, twice daily.
- Children (6-11 years): One spray per nostril, twice daily.
Tablets:
- Adults: One 5 mg tablet twice daily, in the morning and evening. Adjust dosage based on symptoms or age.
- Children (7+ years): One 5 mg tablet twice daily, in the morning and evening.
Note: If a dose is missed, take it as soon as possible unless it’s nearly time for the next dose. Do not double doses.
Administration
Nasal Spray Usage:
- Shake the bottle and remove the dust cover.
- Hold the spray with your fingers on either side of the nozzle and press down until a fine mist appears. For first-time use or if not used for over a week, prime the spray by pressing several times.
- Clear your nostrils by blowing your nose gently.
- Insert the applicator into one nostril, tilt your head slightly forward, and spray while inhaling through your nose.
- Repeat for the other nostril.
Cleaning the Spray:
- Remove the dust cover and nasal applicator.
- Wash both parts in warm water.
- Shake off excess water and let air dry. Avoid heat.
- Reassemble the spray.
Interactions
Olopan Nasal Spray is unlikely to interact with liver enzyme inhibitors, as it is mainly excreted via the kidneys. No significant interactions with P450 enzymes or plasma protein binding are anticipated.
Contraindications
Olopan should not be used by individuals with known hypersensitivity to any of its components. Inform your healthcare provider about your current medications and medical history to ensure safety.
Side Effects
Potential side effects include headaches, dry eyes, burning or stinging sensations, and a range of other symptoms such as nausea, rhinitis, and sinusitis. Rarely, more severe reactions may occur.
Pregnancy & Lactation
