Indications
Nintoin is indicated for the treatment and prevention of urinary tract infections caused by susceptible strains of Escherichia coli, Enterococci, Staphylococcus aureus, Staphylococcus saprophyticus, and certain strains of Klebsiella and Enterobacter species.
Pharmacology
Nitrofurantoin is an antibacterial agent specifically targeting urinary tract infections. It is highly soluble in urine and may impart a brown color. The mechanism of action involves inactivating or altering bacterial ribosomal proteins and other macromolecules, showing efficacy against various Gram-positive and Gram-negative bacteria.
Dosage
Adults:
Uncomplicated UTIs: 50-100 mg four times daily for one week or at least three days post-sterility.
Long-term suppression: 50-100 mg at bedtime.
Capsules:
Acute UTIs: 50 mg four times daily for 7 days.
Long-term suppression: 50-100 mg once daily.
Prophylaxis: 50 mg four times daily during the procedure and for three days after.
Suspension for Children:
Doses based on weight, ranging from 5-7 mg/kg/day in four divided doses.
Administration
Nitrofurantoin should be taken with food.
Interaction
Antacids containing magnesium trisilicate may reduce the absorption of Nintoin.
Contraindications
Contraindicated in patients with anuria or oliguria, significant renal impairment, and known hypersensitivity to Nitrofurantoin.
Side Effects
Common side effects include nausea, headache, and flatulence. Less frequent effects may involve diarrhea, abdominal pain, dizziness, and drowsiness.
Pregnancy & Lactation
Pregnancy Category B; use only if necessary. Nitrofurantoin is found in trace amounts in breast milk.
Precautions & Warnings
Discontinue use if acute pulmonary reactions occur.
Overdose Effects
Symptoms mainly include vomiting; emesis induction is recommended.
