Indications
Loratin Fast tablets swiftly alleviate the symptoms of seasonal allergic rhinitis, perennial allergic rhinitis, and skin allergies like chronic urticaria. They effectively ease manifestations such as sneezing, nasal discharge, itching, ocular itching, and burning associated with allergic rhinitis. These tablets rapidly relieve nasal and ocular signs and symptoms upon oral intake. Moreover, Loratin Fast tablets are indicated for idiopathic urticaria. For children aged 2 and above, Loratin Fast tablets provide symptomatic relief for seasonal allergic rhinitis and allergic skin conditions such as urticaria and nettlerash.
Pharmacology
Loratin Fast tablets contain Loratadine, a non-sedative histamine H1 receptor antagonist with potent antiallergic properties. This long-acting tricyclic antihistamine selectively antagonizes peripheral H1 receptors without inducing central sedation or anticholinergic effects. Its rapid onset and prolonged action enable once-daily administration.
Dosage & Administration
For adults and children aged 12 years and above, one Loratadine tablet is recommended once daily, preferably in the morning or as needed for symptom management. Children aged 2-12 years with a body weight over 30 kg should take one Loratadine tablet daily, whereas those below 30 kg should take half a tablet daily. Loratadine tablets are not recommended for use in children under 2 years due to lack of established safety and efficacy.
Interaction
Loratin Fast tablets have not been reported to interact adversely with other drugs. Unlike many other histamine H1 receptor antagonists, they do not potentiate the effects of alcohol on psychomotor performance. However, concurrent use with drugs metabolized by hepatic cytochromes P450 3A4 and 2D6 may increase plasma concentrations of either drug, potentially leading to adverse effects.
Contraindications
Loratadine is contraindicated in individuals with a known hypersensitivity or idiosyncrasy to its components.
Side Effects
Clinical studies have shown that the incidence of adverse events, including sedation and anticholinergic effects, with 10 mg Loratin Fast is comparable to placebo. Studies assessing its impact on driving performance, cognitive, and psychomotor functioning have demonstrated no significant difference from placebo.
Pregnancy & Lactation
Due to insufficient data on Loratadine use during human pregnancy, its use is not advisable. While Loratadine is excreted in breast milk in minimal amounts, nursing mothers are advised against its consumption.
Precautions & Warnings
Caution should be exercised in patients with liver impairment or renal insufficiency (eGFR <30 ml/min).
Overdose Effects
Overdose may lead to somnolence, tachycardia, and headache in adults, while extrapyramidal signs and palpitations may occur in children. Prompt symptomatic and supportive measures should be initiated in case of overdose, including oral activated charcoal administration or gastric lavage if appropriate.
