Indications
Patadin eye drops are designed to alleviate the signs and symptoms of allergic conjunctivitis, which may include itchy, watery, red, and swollen eyes and/or eyelids. This formulation is effective for conditions such as vernal keratoconjunctivitis, vernal keratitis, blepharitis, blepharoconjunctivitis, and giant papillary conjunctivitis.
Lopadine Nasal Spray, an H1 receptor antagonist, provides relief from seasonal allergic rhinitis symptoms in adults and children aged 6 years and older.
Lopadine tablets are effective in treating allergic rhinitis—an inflammation of the nasal membranes triggered by allergens like pollen and mold spores—as well as allergic skin conditions. They help alleviate symptoms such as skin rashes, hives (itchy, painful eruptions), itching, sneezing, and both runny and stuffy noses.
Pharmacology
Olopatadine, an antihistamine, reduces allergy symptoms by blocking the action of histamine in the body. It effectively treats sneezing, itching, and runny nose associated with allergies.
Mode of Action: Olopatadine serves as an anti-allergic agent through multiple mechanisms. As a mast cell stabilizer, it prevents the release of histamine from human conjunctival mast cells and rodent basophils. Olopatadine selectively antagonizes histamine H1 receptors, blocking their signaling pathway and inhibiting the release of inflammatory mediators like tryptase, prostaglandin D2, TNF-alpha, and pro-inflammatory cytokines.
Pharmacokinetics:
- Absorption: Peak plasma concentrations of olopatadine are reached within 30 minutes to 1 hour after twice-daily administration of the nasal spray.
- Distribution: Approximately 55% of olopatadine binds to serum proteins, regardless of drug concentration.
- Metabolism: Olopatadine is minimally metabolized, with at least six minor metabolites present in the plasma.
- Elimination: The plasma elimination half-life ranges from 8 to 12 hours, with primary excretion occurring through urine.
Dosage
Eye Drops:
- 0.1%: One drop in each affected eye twice daily (6 to 8 hours apart).
- 0.2%: One drop in the affected eye once daily.
- 0.7%: One drop in each affected eye once daily.
Nasal Spray:
- For intranasal use only.
- Adults and Adolescents (12+ years): Two sprays per nostril twice daily.
- Children (6-11 years): One spray per nostril twice daily.
Tablet:
- Adults: One tablet (5 mg) twice daily, in the morning and evening. Dosage may be adjusted based on symptoms or physician recommendations.
- Children (7+ years): One tablet (5 mg) twice daily, in the morning and evening.
- If a dose is missed, take it as soon as remembered. If it’s close to the next dose, skip the missed dose. Do not double doses.

Administration of Nasal Spray:
- Shake the bottle gently and remove the dust cover.
- Hold the spray with your forefinger and middle finger on either side of the nozzle, thumb underneath. Press down until a fine spray is visible. If it’s the first use or hasn’t been used in over a week, press multiple times until a fine mist appears.
- Gently clear nostrils by blowing your nose.
- Close one nostril and insert the nasal applicator into the open nostril. Tilt your head slightly forward, keeping the spray upright. Inhale through your nose while pressing the applicator to release the spray.
- Exhale through your mouth.
- Repeat for the other nostril if necessary.
Cleaning: Clean the nasal spray weekly:
- Remove the dust cover.
- Detach the nasal applicator.
- Wash both parts in warm water.
- Shake off excess water and let them dry naturally, avoiding heat sources.
- Reattach the applicator and dust cover.

Interactions
No formal drug-drug interaction studies have been conducted for Lopadine Nasal Spray. Significant interactions with liver enzyme inhibitors are unlikely, as olopatadine is primarily excreted via the kidneys. Interactions involving P450 inhibition and plasma protein binding are also not anticipated.
Contraindications
This product is contraindicated for individuals with known hypersensitivity to any of its components. Olopatadine tablets are not recommended for those allergic to them. Always inform your healthcare provider about current medications and existing medical conditions to ensure safety.
Side Effects
Headaches are rare, but other potential side effects may include asthenia, blurred vision, stinging or burning sensations, dry eyes, keratitis, lid edema, nausea, pharyngitis, pruritus, sinusitis, and altered taste.
Pregnancy & Lactation
Olopatadine has shown no teratogenic effects in animal studies, but well-controlled studies in pregnant women are lacking. Use during pregnancy should be based on a risk-benefit analysis. It’s unclear whether topical administration results in significant levels in breast milk; caution is advised when prescribing to nursing mothers.
Precautions & Warnings
Patients should avoid wearing contact lenses if experiencing redness in the eyes. Patadin eye drops should not be used for irritation related to contact lenses. The preservative Benzalkonium Chloride in Patadin may be absorbed by soft contact lenses. If you wear soft lenses, wait at least 10 minutes after applying the drops before inserting them. Discontinue use if an allergic reaction occurs.
For initial use of Lopadine Nasal Spray, prime the spray by releasing five sprays until a fine mist appears. If not used for over a week, re-prime with two sprays. Avoid contact with eyes.
Olonex Tablets should be taken as directed by your physician, preferably at the same times each day. Avoid exceeding the recommended dosage and consult your doctor before discontinuing use to prevent symptom recurrence.
Use in Special Populations
Geriatric Use: No significant differences in safety or efficacy have been noted.
Overdose Effects
There’s limited information on Lopadine overdose. However, excessive use of any medication can be serious. If an overdose is suspected, seek immediate medical assistance.
