Pharmacology
Glucosamine: This essential amino sugar, naturally produced by the body and present in some foods, is crucial for synthesizing glycosaminoglycans (GAGs) such as Hyaluronic Acid, Keratan Sulfate, and Chondroitin Sulfate. These GAGs form proteoglycans, the key components of articular cartilage. Supplementing with Glucosamine Sulfate boosts glucosamine levels, aiding in cartilage repair and formation. It also activates chondrocytes, which are vital for producing GAGs and proteoglycans. Absorbed efficiently through the digestive system and metabolized by the liver, glucosamine is mainly excreted through urine.
Diacerein: Specifically used for osteoarthritis, Diacerein offers analgesic, antipyretic, and anti-inflammatory benefits. It inhibits Interleukin-1 (IL-1), a cytokine involved in cartilage breakdown, thereby demonstrating disease-modifying effects in both experimental and human studies. Diacerein is best taken with food to enhance absorption and minimize delays in peak concentration. It binds primarily to albumin, is extensively metabolized in the liver, and its metabolites are excreted through urine.
Dosage & Administration
- Adults: One tablet twice daily with food.
- Children and Adolescents: Safety and efficacy for individuals under 18 years have not been established. Follow medical advice for dosage and administration.
Interaction
- Decreased Absorption: Avoid combining Diacerein with antacids containing aluminum or magnesium hydroxide.
- Increased Risk of Diarrhea: Use with caution alongside laxatives or antibiotics.
Contraindications
Not recommended for individuals with hypersensitivity to Glucosamine or Diacerein.
Side Effects
Generally well-tolerated; possible side effects include nausea, vomiting, diarrhea, abdominal pain, headache, skin rashes, and dark yellow urine.
Pregnancy & Lactation
Jointec Max is contraindicated during pregnancy and breastfeeding.
