Pharmacology
Glucosamine: Glucosamine (2-amino-2-deoxy-alpha-D-glucose) is a natural amino sugar produced by the body and found in various foods. It is a vital building block for the biosynthesis of glycosaminoglycans (GAGs) such as hyaluronic acid, keratan sulfate, and chondroitin sulfate. GAGs combine with proteins to form proteoglycans, essential components of articular cartilage. When cartilage deteriorates, osteoarthritis can develop. Glucosamine also aids in the formation of ligaments, tendons, nails, and other connective tissues. Supplementing with synthesized glucosamine sulfate elevates glucosamine levels in the body, promoting cartilage production and repair. Additionally, glucosamine activates chondrocytes within cartilage, stimulating the production of GAGs and proteoglycans.
Approximately 90% of glucosamine administered as an oral dose of glucosamine sulfate is absorbed through the digestive system, primarily metabolized by the liver, and excreted via urine.
Diacerein: Diacerein is indicated for treating osteoarthritis and possesses analgesic, antipyretic, and anti-inflammatory properties. It inhibits interleukin-1 (IL-1) synthesis in vitro, a key cytokine involved in cartilage degradation. Due to its specific mechanism, diacerein has shown disease-modifying effects in experimental osteoarthritis models and in human subjects with finger joint and knee osteoarthritis.
Diacerein has an oral bioavailability of 56%. Taking it with food delays peak concentration but increases absorption by 25%, making it advisable to administer diacerein with meals. It primarily binds to the protein albumin and is extensively metabolized in the liver following oral dosing, with urinary excretion of its metabolites ranging between 35% and 60%.

Interactions
Diacerein absorption may be reduced when taken with aluminum and/or magnesium hydroxide antacids. Concurrent use with laxatives or antibiotics may increase the risk of diarrhea.
Contraindications
Joinix D is contraindicated for individuals with hypersensitivity to glucosamine or diacerein.
Side Effects
Serious adverse effects have not been reported. However, some individuals may experience nausea, vomiting, diarrhea, epigastric pain, headaches, skin rashes, and intense yellow urine.
Pregnancy & Lactation
This tablet is contraindicated during pregnancy and breastfeeding.
Precautions & Warnings
Use caution when administering this tablet to patients with known allergies to glucosamine or diacerein.
