Pharmacology of Gabarol
Pregabalin, the active ingredient in Gabarol, is a synthetic derivative of gamma-aminobutyric acid (GABA). Although it does not directly bind to GABAA, GABAB, or benzodiazepine receptors, Pregabalin works by attaching to the alpha2-delta subunit of voltage-gated calcium channels in the central nervous system. This mechanism is thought to contribute to its effectiveness in treating neuropathic pain and managing seizures.
Dosage Guidelines
- For Diabetic Peripheral Neuropathy (DPN): Start with 50 mg three times a day (150 mg/day), with a gradual increase up to 300 mg/day based on effectiveness and tolerance. For Gabarol CR, begin with 165 mg once daily, increasing to 330 mg within one week.
- For Postherpetic Neuralgia: Start with 75 mg twice a day or 50 mg three times a day, with a gradual increase to 300 mg/day. In case of insufficient pain relief, increase the dose to 600 mg/day.
- For Fibromyalgia: Begin with 75 mg twice a day (150 mg/day), gradually increasing to 450 mg/day.
- For Neuropathic Pain with Spinal Cord Injury: Start with 75 mg twice a day (150 mg/day), and increase up to 600 mg/day based on individual response.
Dosing adjustments are based on individual needs and should be done under medical supervision.
Administration of Gabarol
- Oral Administration: Gabarol can be taken with or without food. The controlled-release (CR) formulation should be taken after an evening meal. Do not chew, crush, or split the tablets. If a dose is missed, follow the instructions carefully depending on the timing of the next dose.
- Gradual Discontinuation: When stopping Gabarol, reduce the dosage gradually over a period of at least one week to prevent withdrawal symptoms.
Drug Interactions
- Gabarol is unlikely to cause significant pharmacokinetic interactions with other medications.
- Always inform your healthcare provider about all other medications you are taking to avoid potential interactions.
Contraindications and Warnings

Precautions
- Respiratory depression may occur if Gabarol is combined with other CNS depressants.
