Indications
Afun VT rectal preparation effectively addresses vaginal itching, burning, and discharge associated with recurrent vaginal yeast infections (Vaginal candidiasis) and vaginitis due to Candida species, including Mycotic infections complicated by other microorganisms sensitive to the drug.
Pharmacology
Clotrimazole, a broad-spectrum antifungal agent, combats dermal infections caused by various pathogenic dermatophytes, yeasts, molds, and bacteroids. It also exhibits activity against certain gram-positive bacteria, especially Staphylococci, and Trichomonas. Clotrimazole alters fungal cell wall permeability by interfering with cell wall lipids, inhibiting ergosterol synthesis at fungistatic levels, and causing membrane damage at higher concentrations, unrelated to sterol synthesis.
Dosage
For initial infections, a 3-day treatment regimen with Clotrimazole 200 mg vaginal preparation is generally effective for Candida vaginitis. For re-infections or mixed infections, a 6-day treatment plan is recommended. Clotrimazole 100 mg vaginal preparation can be used nightly for six consecutive nights. Additionally, Clotrimazole 500 mg vaginal preparation offers a single-dose, complete treatment. Partners should undergo local treatment with Clotrimazole cream simultaneously to prevent re-infection.
Administration
To administer Clotrimazole vaginal preparation, pull out the plunger until it stops, place a vaginal tablet into the applicator, carefully insert the applicator containing the tablet as deeply as possible into the vagina while lying on your back, push the plunger until it stops to deposit the tablet, then remove the applicator. After use, clean the applicator thoroughly. Pregnant women should follow their physician’s instructions.

Side Effects
Due to minimal absorption through vaginal skin, systemic effects of Afun VT are unlikely. Local tolerance is generally good, with rare instances of local irritation or burning, which are considered non-harmful.
Pregnancy & Lactation
Clotrimazole should be used during pregnancy only when deemed necessary by a physician. Studies indicate minimal absorption into the bloodstream and no adverse effects in women in their second or third trimesters. Its secretion in breast milk is unknown. Clotrimazole suppositories can be used during pregnancy under physician supervision.
