Delanix 60

Indications

Delanix is prescribed for:

• Healing Erosive Esophagitis (EE): Effective treatment for all grades of EE for up to 8 weeks.
• Maintenance of Healed EE: Helps maintain healing and alleviate heartburn for up to 6 months.
• Symptomatic Non-Erosive Gastroesophageal Reflux Disease (GERD): Treats heartburn associated with non-erosive GERD for 4 weeks.

Pharmacology

Delanix contains Dexlansoprazole, a Proton Pump Inhibitor (PPI) that effectively reduces gastric acid secretion by specifically inhibiting the (H+/K+)-ATPase in gastric parietal cells. This action blocks the final stage of acid production. The Dual Delayed Release (DDR) formulation leads to two distinct plasma concentration peaks: the first peak occurs 1 to 2 hours post-administration, followed by a second peak within 4 to 5 hours. Dexlansoprazole is primarily metabolized in the liver and excreted through urine.

Dosage

Dexlansoprazole Dosing Guidelines:

• Maintenance of Healed EE: 30 mg once daily.
• Symptomatic Non-Erosive GERD: 30 mg once daily for 4 weeks.
• Healing of Erosive Esophagitis: 60 mg once daily for up to 8 weeks.

MUPS Dosing Recommendations:

• Maintenance of Healed EE: 30 mg tablet once daily for 6 months (adults) and 16 weeks (ages 12 to 17).
• Symptomatic Non-Erosive GERD: 30 mg tablet once daily for 4 weeks.

Administration

Delanix can be taken without regard to meals and should be swallowed whole. Alternatively, capsules can be opened, and granules sprinkled on one tablespoon of food for immediate consumption. Do not chew the granules. If a dose is missed, take it as soon as possible unless it’s close to the next dose; do not double the dose.

Interactions

Caution is advised when using Delanix with medications such as Atazanavir, Warfarin, Tacrolimus, Clopidogrel, and Methotrexate.

Contraindications

Delanix is contraindicated for individuals with known hypersensitivity to any of its components.

Side Effects

Common side effects may include diarrhea, abdominal pain, nausea, vomiting, and flatulence.

Pregnancy & Lactation

Classified as Pregnancy Category B; there are no well-controlled studies in pregnant or lactating women.

Precautions & Warnings

Use Delanix with caution in cases of gastric malignancy, Clostridium difficile-associated diarrhea, risk of bone fractures, hypomagnesemia, and alongside Methotrexate.

Use in Special Populations

• Children & Adolescents: Safety and efficacy have not been established for those under 12 years.
• Geriatric Use: No dose adjustment required for elderly patients.
• Renal & Hepatic Impairment: No adjustments needed for mild renal impairment. For moderate hepatic impairment, a maximum dose of 30 mg is recommended.

Overdose Effects

No significant overdose reports have been recorded. Doses up to 300 mg have not resulted in severe adverse events.
