Pharmacological Profile: Glucosamine
Glucosamine (2-amino-2-deoxy-alpha-D-glucose) is a natural amino sugar, naturally synthesized by the body and present in certain foods. It acts as the primary building block crucial for the biosynthesis of glycosaminoglycans (GAGs) such as Hyaluronic Acid, Keratan Sulfate, and Chondroitin Sulfate. These GAGs combine with proteins to form proteoglycans, which are vital for the construction of articular cartilage. Osteoarthritis develops when cartilage in a joint deteriorates. Glucosamine supplementation, particularly in the form of artificially synthesized Glucosamine Sulfate, elevates Glucosamine levels in the body, thus promoting cartilage production and repair. Additionally, Glucosamine activates chondrocytes within the cartilage, stimulating the production of GAGs and proteoglycans.
Approximately 90% of orally administered glucosamine sulfate is absorbed from the digestive tract in humans. It is primarily metabolized by the liver and excreted through urine.
Diacerein
This is utilized in treating Osteoarthritis and possesses analgesic, antipyretic, and anti-inflammatory properties. It inhibits Interleukin-1 (IL-1) synthesis directly, a key cytokine involved in cartilage degradation. Due to its specific mode of action, it has demonstrated disease-modifying effects in experimental osteoarthritis models and in human subjects with finger joint and knee osteoarthritis.
Diacerein exhibits an oral bioavailability of 56%. Concurrent intake with food delays the time to reach peak concentration but is associated with a 25% increase in absorption. Hence, administering diacerein with food is optimal. It predominantly binds to the protein albumin. After oral dosing, diacerein undergoes extensive (100%) metabolism in the liver. Urinary excretion of diacerein in the form of its metabolites ranges between 35% and 60%.
Dosage & Administration
For adults: Take one tablet twice daily with food.
For children and adolescents: The safety and efficacy in individuals under 18 years old have not been established.

Interactions
Aluminum and/or magnesium hydroxide antacids reduce the absorption of Diacerein. There is an increased risk of diarrhea when administered with laxatives or antibiotics.
Contraindications
Contraindicated for individuals exhibiting hypersensitivity to Glucosamine or Diacerein.
Side Effects
No serious adverse effects have been reported. Potential side effects may include nausea, vomiting, diarrhea, epigastric pain, headache, skin rashes, and intense yellow coloring of urine.
Pregnancy & Lactation
This tablet is contraindicated during pregnancy and breastfeeding.
